چکیده

بارانک ایرانی (Sorbus persica Hedl.) درختی کُند رشد از تیره رزاسه و از گونه‌های انحصاری ایران و در حال انقراض است. از لحاظ ذخیره ژنی، حفظ زیست محیطی و کاربرد دارویی حائز اهمیت است. به منظور تکثیر درون‌شیشه‌ای این گونه به روش‌ کشت جوانه جانبی، پژوهشی به‌صورت فاکتوریل بر پایه طرح کاملاً تصادفی در سه سطح محیط کشت و سه سطح سیتوکینین در سه تکرار اجرا شد. برای شاخه‌زایی، محیط‌های‌کشت ‌DKW، WPM و MS حاوی سیتوکینین‌های BAP، Kin و 2ip و برای ریشه‌زایی، محیط‌های کشت ‌DKW، WPM و MS کامل و تغییر یافته با اکسین‌های IBA و NAA به‌صورت مجزا و کاربرد متوالی بررسی شدند. نتایج نشان داد که بهترین تیمار سترون‌سازی، کاربرد متوالی محلول هیپوکلریت سدیم 20% (حجمی/حجمی، 15 دقیقه) و کلرید جیوه 0.1% (3 دقیقه) در فصل تابستان بود. تکثیر بهینه و رشد طولی شاخساره‌ها در محیط کشت MS با تنظیم‌کننده‌های رشد IBA (0.01 میلی‌گرم در لیتر)، Kin (0.25 میلی‌گرم در لیتر) و BAP (3 میلی‌گرم در لیتر) مشاهده شد. همچنین، ریشه‌زایی بهینه در محیط کشت DKW با یک چهارم غلظت عناصر بیش مغذی حاوی هورمون‌های NAA+IBA (0.3+0.3 میلی‌گرم در لیتر) بدست آمد.
