چکیده

این مطالعه به‌منظور بررسی تأثیر بیوپرایمینگ با تعدادی از جدایه‌های بومی قارچ Trichoderma harzianum روی شاخص‌های جوانه‌زنی و بنیه بذر یک توده بومی زیره سبز (Cuminum cyminum L.) انجام شد. همچنین تأثیر آنزیم‌های خارج سلولی تولیدشده توسط این جدایه‌ها به‌عنوان الیسیتور بر تولید و انباشت متابولیت‌های ثانویه در گیاهچه‌های حاصل از بذرهای بیوپرایم شده بررسی شد. براساس نتایج، تمام جدایه‌های T. harzianumقادر به تولید آنزیم‌های آمیلاز، پروتئاز، سلولاز، زایلاناز، کیتیناز و لیپاز بودند. نتایج همچنین نشان داد که بیوپرایمینگ بذر زیره سبز به‌طور قابل‌توجهی روی شاخص‌های بنیه و جوانه‌زنی بذر تأثیر گذاشته و موجب بهبود کیفیت و سلامت بذر و گیاهچه شد. در این مطالعه، اسانس گیاهچه‌های حاصل از بذرهای بیوپرایم شده به روش تقطیر با آب استخراج و ترکیبات شیمیایی آن با استفاده از روش‌های کروماتوگرافی گازی و کروماتوگرافی گازی متصل به طیف‌سنج جرمی شناسایی شدند. نتایج نشان داد که ترکیب‌های اصلی شناسایی شده شامل بتا-پینن، پارا-سیمن، لیمونن، گاما-ترپینن، ترپینن-4-اُل، آلفا-ترپینئول، کومین آلدهید و بتا-فارنزن بودند. همچنین قارچ T. harzianumو آنزیم‌های ترشح شده توسط آن به‌عنوان الیسیتور موجب افزایش بیان ژن‌های مرتبط با تولید و انباشت متابولیت‌های ثانویه در زیره سبز شدند. این اولین گزارش در مورد تأثیر بیوپرایمینگ بذر با جدایه‌های بومی قارچ T. harzianum روی ترکیب‌های اسانس گیاهچه‌های زیره سبز می‌باشد. یافته‌های این پژوهش نشان داد که مقدار آنزیم‌های خارج سلولی ترشح شده توسط جدایه‌های T. harzianum متفاوت است و روی میزان تولید و انباشت متابولیت‌های ثانویه در زیره سبز تأثیر می‌گذارد.
