DXS gene in the MEP pathway produces 1-deoxy-d-xylulose 5-phosphate using pyruvate and glyceraldehyde-3-phosphate dehydrogenase. TcGLIP gene is involved at the last steps of pyrethrins biosynthesis pathway and integrates chrysanthemyl-CoA and pyrethrolone, which are produced in the MEP pathway and lipoxygenase pathways, respectively to yield pyrethrin I. Based on the significances of pyrethrins, further identification and studies are needed on these metabolites. The aim of this study was to isolate DXS gene in pyrethrum (Chrysanthemum cinerariaefolium Vis.) from the Asteraceae, as well as expression analysis of these two genes under methyl-jasmonate treatment. In the present work, the nucleotide sequences of 1-deoxy-D-xylulose 5-phosphate synthase gene of different plant species were obtained from NCBI database and subsequently aligned with ClustalW online software. Specific primers were designed from the conserved regions of aligned sequences. The polymerase chain reaction product with 715bp length showed that the specific primers had high efficiency to isolate DXS. In addition, gene expression analysis under methyl-jasmonate treatment showed that both DXS and TcGLIP were up-regulated in response to methyl jasmonate elicitor. Our data showed that methyl jasmonate treatment in seedling stage could be used as a suitable elicitor to increase pyrethrin production.}, keywords = {1-deoxy-D-xylulose 5-phosphate synthase,pyrethrum,TcGLIP multifunctional gene,gene isolation,methyl-jasmonate}, title_fa = {بررسی بیان ژن 1-دئوکسی-دی-زایلوز 5-فسفات سنتاز و ژن چند عملکردی TcGLIP در گیاه پیرتروم (Chrysanthemum cinerariaefolium Vis.) در پاسخ به متیل جاسمونات}, abstract_fa = {ژن 1-دئوکسی-دی-زایلوز 5-فسفات سنتاز و ژن چند عملکردی GLIP (TcGLIP) دو ژن کلیدی در مسیر بیوسنتزی "پیرترین‌ها" می‌باشند که دارای خاصیت حشره‌کشی می‌باشند. ژن DXS در مسیر MEP با استفاده از پیروات و گلیسر آلدهید 3-فسفات باعث تولید 1-دئوکسی-دی-زایلوز 5-فسفات می‌گردد و ژن TcGLIP نیز در انتهای مسیر بیوسنتزی دخالت می‌کند و مولکول‌های کریزانتمیل کوآ (Chrysanthemyl CoA) و پیرترلون (Pyrethrolone) توسط دو مسیر MEP و لیپوکسی ژناز را به یکدیگر متصل می‌کند و باعث تولید پیرترین نوع 1 می‌شود. با توجه به اهمیت بالای پیرترین‌ها، بررسی و شناخت بیشتر این متابولیت‌ها حائز اهمیت می‌باشد. هدف از این مطالعه، جداسازی ژن DXS در گیاه پیرتروم (Chrysanthemum cinerariaefolium Vis.) از خانواده گل‌ستاره‌ای‌ها و بررسی بیان دو ژن DXS و TcGLIP تحت تیمار متیل جاسمونات بود. در این تحقیق ابتدا توالی‌های نوکلئوتیدی ژن 1-دئوکسی-دی-زایلوز 5-فسفات سنتاز گونه‌های گیاهی مختلف از پایگاه اطلاعاتی NCBI دریافت شد و بعد توالی‌های دریافت شده توسط نرم‌افزار تحت وب ClustalW هم‌ردیف شدند. از هم‌ردیف‌سازی توالی‌های بدست آمده برای این توالی در گیاه پیرتروم آغازگرهای ویژه طراحی و سنتز شد. نتایج واکنش زنجیره‌ای پلیمراز و نتایج توالی‌یابی محصول آن و مشاهده باند با طول bp715 نشان داد که آغازگرهای اختصاصی طراحی شده برای جداسازی ژن DXS کارایی لازم را دارند. همچنین بررسی بیان این دو ژن تحت تیمار متیل جاسمونات، القای بیان این ژن‌ها را تحت تأثیر متیل جاسمونات در مرحله گیاهچه‌ای نشان داد که نشان می‌دهد که متیل جاسمونات می‌تواند به‌عنوان یک الیسیتور مناسب برای افزایش میزان تولید پیرترین‌ها بکار گرفته شود.}, keywords_fa = {1-دئوکسی-دی-زایلوز 5-فسفات سنتاز,پیرتروم,ژن چند عملکردی TcGLIP,جداسازی ژن,متیل جاسمونات}, url = {https://ijmapr.areeo.ac.ir/article_109703.html}, eprint = {https://ijmapr.areeo.ac.ir/article_109703_2341491b1494b0edcbd108c4e3129715.pdf} }
