چکیده

زعفران خوراکی (Crocus Sativus L.) دارای خواص بیولوژیکی متفاوتی از جمله خاصیت ضدسرطانی است. در این تحقیق، علاوه بر تخلیص مواد مؤثر زعفران، دو سازوکار  عمدة ملکولی در ارتباط با این خاصیت زعفران یعنی اثر بر DNA، هیستون و کمپلکس DNA –H1 مورد بررسی قرار گرفت. کاروتنوئیدها (کروسین، کروستین و دی‌متیل‌کروستین) و آلدئیدهای منوترپن (پیکروکروسین و سافرانال) موجود در زعفران ایران به عنوان اجزاء اصلی، توسط روش‌های مختلفی جداسازی و خالص‌سازی شد. نتایج TLC و HPLC نشان داد که کروسین و پیکروکروسین موجود در زعفران ایران به ترتیب از 4 و 3 زیرنوع تشکیل شده است. بنابراین، جزء اصلی و عمدة کروسین‌ها و پیکروکروسین‌ها تخلیص شد و میان‌کنش هیستون H1 و DNA نه تنها با عصارة زعفران، بلکه با هریک از پنج جزء تخلیص شده از زعفران توسط روشهای مختلف مورد بررسی قرار گرفت. در بررسی میان‌کنش زعفران و اجزاء آن با DNA مشاهده شد که اتصال تمام این لیگاندها از طریق اتصال به شیار کوچک صورت می‌گیرد و منجر به القاء تغییرات ساختاری در  DNAمی‌شود. همچنین در بررسی اثر این لیگاندها بر ساختار هیستون H1 مشاهده شد که این لیگاندها باعث فرونشانی نشر فلورسانس هیستون می‌شوند. با رسم نمودار اسکاچارد براساس فرونشانی نشر، مولفه های پیوندی n وK محاسبه گردید (n و K به ترتیب برابر 54/0 و5-10*2 بدست آمد). طیف دورنگ نمایی دورانی (CD) نشان داد که این ترکیبهای در غلظت کم با DNA میان‌کنش دارند که موجب تغییر ساختار از B-DNA به C-DNA می‌شود، هر چند که در غلظت‌های بالاتر، تغییر ساختاری بیشتری القاء می‌گردد،  یعنی رج بندی بازها از بین می‌رود. همچنین نتایج حاصل از بررسی میان‌کنش DNA –H1 در حضور و عدم حضور هر کدام از لیگاندهای فوق نشان داد که در حضور این اجزاء، میان‌کنش H1 با DNA حدود 5 تا 22 درصد کاهش یافت که بیشترین میزان کاهش، مربوط به سافرانال بود.
