اثر عصاره هیدروالکلی سرخهولیک (Crataegus monogyna Jacq.) بر هیستوپاتولوژی و نشانگرهای آسیب کبدی در موشهای صحرایی دیابتی نر بالغ
شناسه دیجیتال (DOR): 98.1000/1735-0905.1398.35.539.96.4.1575.1594 بیماری دیابت ملیتوس با تغییرات فیزیکی، فیزیولوژیکی و پاتولوژیکی در بافت کبد همراه است. هدف از این مطالعه بررسی اثرهای حفاظتی عصاره هیدروالکلی سرخهولیک (Crataegus monogyna Jacq.) بر هیستوپاتولوژی و عملکرد کبد در موشهای صحرایی دیابتی شده توسط استرپتوزوتوسین بود. در این مطالعه تجربی موشهای صحرایی نر نژاد wistar بهطور تصادفی در 6 گروه 8 تایی (8n=) شامل کنترل، دیابتی درمان نشده، دیابتی درمان شده با دوزهای 100، 200 و 400 (mg/kg) عصاره سرخهولیک و کنترل مثبت (دیابتی درمان شده با کاپتوپریل) تقسیم شدند. پس از 3 هفته تجویز عصاره و دارو بهصورت درون صفاقی، سطح سرمی آنزیمهای کبدی و آلبومین تعیین گردید و نمونه بافت کبد برای بررسیهای هیستوپاتولوژی تهیه شد. برای القاء دیابت mg/kg60 استرپتوزوتوسین (STZ) بهصورت داخل صفاقی تزریق شد و پس از مشاهده علائم دیابت (قند خون بالای mg/dL25۰ و پرنوشی) تیمار توسط عصاره و دارو انجام گردید. در موشهای دیابتی در مقایسه با موشهای کنترل، آلبومین سرم کاهش معنیدار و میزان گلوتامیک پیروویک ترانس آمیناز (GPT) و گاما گلوتامیل ترانسفراز (GGT) سرم افزایش معنیدار نشان دادند (0.05>P). تیمار موشهای دیابتی با دوزهای مختلف عصاره سرخهولیک سبب افزایش معنیدار آلبومین سرم و کاهش معنیدار GGT سرم شد (0.05>P). در موشهای دیابتی نکروز هپاتوسیتها، تخریب کامل لبولها و طنابهای رماک و پرخونی نسبتاً شدید مشاهده شد و تیمار با عصاره سرخهولیک سبب پیشگیری از تغییرات پاتولوژیک گردید. نتایج حاصل از این مطالعه نشان داد عصاره سرخهولیک میتواند سبب پیشگیری از اختلال عملکرد و آسیب بافتی کبد در موشهای صحرایی دیابتی گردد.

بهینهسازی بستر کشت برای استقرار گیاهچه زیره سیاه (Bunium persicum (Boiss.) B. Fedttsch.)
شناسه دیجیتال (DOR): 98.1000/1735-0905.1398.35.551.96.4.1576.41 اهلیسازی، کاشت و تولید زراعی زیره سیاه (Bunium persicum (Boiss.) B. Fedttsch.) برای جلوگیری از کاهش ذخایر ژنتیکی آن از اهمیت ویژهای برخوردار است. در همین راستا آزمایشی با هدف شناسایی بستر و برنامه غذایی مناسب برای استقرار گیاهچه یکساله زیره سیاه بهصورت فاکتوریل در قالب طرح پایه کاملاً تصادفی در شرایط گلخانهای با سه تکرار اجرا شد. عوامل آزمایش شامل سه بستر کوکوپیت، ماسه و خاک زراعی و پنج غلظت محلول هوگلند (صفر (بدون محلول غذایی)، 25، 50، 75 و 100 درصد) بود. نتایج نشان داد که بیشترین درصد سبز (37.67) در بستر کوکوپیت و بدون محلول غذایی حاصل شد. بیشترین شاخص بنیه گیاهچه (773) و سطح سبز (2.14 سانتیمترمربع در بوته) در بستر کوکوپیت و تغذیه با هوگلند 75% و کمترین شاخص بنیه گیاهچه (205) در بستر خاک و تغذیه با هوگلند 75% مشاهده شد. بالاترین درصد غدهدهی (100) در بستر ماسه و تغذیه با محلول هوگلند 50% بدست آمد. بزرگترین غدهها از نظر وزن (76.0 میلیگرم) و حجم (79.2 میلیمترمکعب) در تیمار بستر ماسه و تغذیه با محلول غذایی هوگلند 75% بدست آمد که با تیمار بستر ماسه و تغذیه با محلول هوگلند 50% تفاوت معنیداری نداشت. بهطور کلی انتخاب بستر با تخلخل و ظرفیت نگهداری آب مناسب و تغذیه با محلول هوگلند حداقل 50% در رسیدن به درصد سبز و استقرار بالای گیاهچه زیره سیاه و تولید غده با اندازه مناسب میتواند مؤثر باشد.

اثرهای زیستی عصارههای آقطی، گزنه، تمشک و سرخس عقابی روی کنه تارتن دو نقطهای
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.565.96.4.1575.1610 بررسی کارایی ترکیبهای گیاهی برای کنترل آفات، یکی از پژوهشهای رو به گسترش در بیشتر سیستمهای کشاورزی است. در این پژوهش کارایی عصاره آبی و اتانولی چهار گیاه آقطی (Sambucus ebulus L.)، گزنه (Urtica dioica L.)، تمشک (Rubus fruticosus Boiss.) و سرخس عقابی (Pteridium aquilinum (L.) Kuhn) روی مادههای بالغ کنه تارتن دو نقطهای (Tetranychus urticae) که یکی از مهمترین آفتهای محصولات کشاورزی و گلخانهای میباشد، بررسی شد. آزمایشها شامل اثر کشندگی و دوام آن و اثر بازدارندگی تخمگذاری بودند. عصاره آبی تمشک اثر کشندگی روی این آفت نداشت و میزان مرگ و میر ناشی از عصاره آبی سه گیاه دیگر از 9.2 تا 49.8 درصد متغیر بود. مقدار LC50 عصارههای اتانولی آقطی، گزنه، سرخس عقابی و تمشک برای کنههای ماده بالغ بهترتیب 0.54، 0.82، 0.72 و mg/cm2 0.76 برآورد شد. در بررسی دوام کشندگی عصارههای اتانولی مشخص شد که عصاره گزنه تا 48 ساعت دوام بالایی داشت، بعد از آن بهترتیب عصارههای تمشک، آقطی و سرخس عقابی قرار گرفتند. کاهش تخمگذاری در کنههای تیمار شده با عصاره اتانولی تمشک (92.98% در بالاترین غلظت) بیش از سایر عصارهها بود. نتایج این تحقیق نشان داد که عصارههای اتانولی کارایی مناسبی داشته و بررسی اثر کنترلی آنها در شرایط مزرعهای، دادههای کاربردی باارزشی در اختیار خواهد گذاشت.

اثر نوع ریزنمونه بر مقدار ماده مؤثره تولید شده در شرایط In vitro در دو گونه Betula pendula Roth و Betula litwinowii
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.577.96.4.1575.105 بتولین و اسید بتولینیک یکی از مهمترین متابولیتهای ضد سرطان (پوست، سینه، رحم، روده، پانکراس، مغز استخوان و غیره) و ضد ایدز میباشند که منبع اولیه تهیه آن درخت توس است. با توجه به در حال انقراض بودن این درختان در ایران، جایگزین نمودن روشهای نوین کشت سلول و بافت بجای استخراج از پوست امری ضروری میباشد. هدف از انجام این مطالعه تعیین میزان تأثیر نوع ریزنمونه بر مقدار بتولین و بتولینیک اسید تولید شده در کالوسهای دو گونه Betula pendula و B. litwinowii در مقایسه با نمونههای گرفته شده از طبیعت بوده است. ریزنمونه برگ و پوست گونههای مذکور با هدف کالزایی در محیط کشت پایه WPM حاوی 1 میلیگرم بر لیتر BAP و 0.1 میلیگرم بر لیتر 2,4-D کشت شدند. میزان بتولین و اسید بتولینیک کالوسهای سه ماهه با استفاده از تکنیک HPLC ارزیابی و با میزان موجود در ساقههای یک سانتیمتری دو گونه (نمونههای گرفته شده از طبیعت) مقایسه شد. ریزنمونه پوست بهمراتب موفقتر از ریزنمونه برگ در کالزایی عمل نمود و در عین حال میزان ماده مؤثره بیشتری نیز در آن القاء شد. میزان بتولین و اسید بتولینیک استخراجی از عصاره پوست طبیعی B. pendula بهترتیب 5.227 و 2.909 درصد و B. litwinowii بهترتیب 649/5 و 519/2 درصد بود. همچنین کالوسهای حاصل از پوست B. pendula و B. litwinowi بهترتیب دارای 0.0226 و 0.016 درصد بتولین و 0.0525 و 0.057 درصد اسید بتولینیک بودند. بهطور کلی ریزنمونه پوست در هر دو گونه توس، باعث القاء مقدار بیشتری ماده مؤثره نسبت به ریزنمونه برگی در شرایط In vitro شد.

بررسی اثر عصاره چند گیاه دارویی بر مهار قارچ Alternaria solani عامل بیماری لکهموجی گوجهفرنگی
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.588.96.4.1576.1610 بیماری لکهموجی ناشی از قارچ Alternaia solani یکی از مهمترین بیماریهای گوجهفرنگی در ایران و سایر نقاط جهان میباشد. بهمنظور بررسی فعالیت ضد قارچی چند گیاه دارویی برای کنترل قارچ Alternaria solani در شرایط درون شیشه و گلخانه آزمایشهایی مجزا در قالب طرح کاملاً تصادفی اجرا شد. در آزمایش اول اثر غلظتهای 200، 400 و ppm600 عصاره آبی و الکلی آویشن زوفایی (Thymbra spicata L.)، آویشن کرکآلود (Thymus eriocalyx (Ronniger) Jalas)، مرزه خوزستانی (Satureja khuzistanica Jamzad) و لعل کوهستان (Oliveria decumbens Vent.) همراه با شاهد (آب مقطر) در شرایط درون شیشه مورد بررسی قرار گرفت که نتایج حکایت از مهار کامل رشد پرگنه قارچ در کلیه تیمارها داشت. در آزمایش دوم 72 ساعت پس از آلودهسازی بوتههای گوجهفرنگی با عامل بیماری لکهموجی در داخل گلخانه، محلولپاشی با غلظتهای 200، 400 و ppm600 عصارههای مذکور اعمال و 14 روز بعد مورد ارزیابی قرار گرفتند. در آزمایش سوم نیز اثر حفاظتی تیمارهای فوق مورد مطالعه قرار گرفت. نتایج آزمایشهای دوم و سوم نشان داد که با افزایش غلظت عصاره، اثر مهارکنندگی و حفاظتی تیمارها نیز افزایش یافت. همچنین، اگرچه همه تیمارها باعث کاهش درصد شدت بیماری نسبت به شاهد (آب مقطر) شدند، ولی غلظت ppm600 عصاره آویشن زوفایی، ppm600 آویشن کرکآلود و ppm400 آویشن زوفایی بهترتیب بیشترین اثر مهارکنندگی و حفاظتی بر رشد قارچ مورد مطالعه داشتند. بنابراین، استفاده از تیمارهای فوق بهعنوان برنامهای کاربردی برای کنترل بیولوژیک قارچ Alternaria solani عامل بیماری لکهموجی گوجهفرنگی توصیه میگردد.

اثر نیتریک اکسید بر فیزیولوژی و متابولیتهای ثانویه گیاه دارویی ریحان (Ocimum basilicum L.) در شرایط تنش خشکی
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.601.96.4.1575.1610 تنش خشکی بهعنوان یکی از مهمترین تنش غیرزیستی نقش زیادی در کاهش عملکرد گیاهان دارویی دارد. بهمنظور بررسی تأثیر نیتریک اکسید بر خصوصیات مورفوفیزیولوژیک و بیوشیمیایی گیاه ریحان (Ocimum basilicum L.) توده بومی شهرری در شرایط تنش خشکی آزمایشی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار انجام شد. فاکتورهای مورد آزمایش شامل تنش خشکی (شاهد (آبیاری کامل) و قطع آبیاری در مرحله ابتدای گلدهی و 50% گلدهی) و محلولپاشی نیتریک اکسید بهصورت سدیم نیتروپروساید در غلظتهای 0.5، 1 و 2 میلیمولار بودند. در تیمار شاهد نیز گیاهان با آب مقطر اسپری برگی شدند. نتایج نشان داد با افزایش شدت تنش خشکی ارتفاع بوته، وزن خشک گیاه، کلروفیل a، کلروفیل b، کلروفیل کل و کاروتنوئید و محتوای آب نسبی کاهش معنیداری یافت و میزان نشت الکترولیت، محتوای پرولین برگ و درصد اسانس افزایش یافت. محلولپاشی با نیتریک اکسید، باعث افزایش رشد گیاه ریحان در شرایط تنش خشکی شد و بیشترین بهبود رشد در اثر محلولپاشی غلظت 2 میلیمولار از نیتریک اکسید حاصل گردید. با افزایش تنش خشکی و غلظت نیتریک اکسید مقدار ترکیبهای اسانس افزایش یافت. عمدهترین ترکیبهای اسانس ریحان در این آزمایش شامل متیل کاویکول، لینالول، ژرانیال، ژرانیول، آلفا-پینن، بتا-پینن و میرسن بودند. بهطور کلی نتایج این آزمایش نشان داد که محلولپاشی نیتریک اکسید در شرایط تنش خشکی با ایجاد تنظیم اسمزی، جلوگیری از تجزیه کلروفیل و حفظ محتوای آب برگ در بهبود رشد گیاه ریحان مؤثر میباشد.

شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.617.96.4.1575.101 انسداد آوندها بهدلیل رشد عوامل میکروبی بهویژه باکتریها، یکی از مهمترین دلایل کاهش کیفیت و عمر گلجایی گلهای بریدنی در مرحله پس از برداشت است. بهمنظور بررسی اثر عصارههای 1% و 2% و همچنین اسانسهای ppm25 و ppm50 آویشن زوفایی (Thymbra spicata L.) و مرزه بختیاری (Satureja bachtiarica L.) بر جمعیت باکتریایی و عمر گلجایی ژربرا (Gerbera jamesoni L.) رقم Pink Elegance آزمایشی بهصورت فاکتوریل بر پایه طرح کاملاً تصادفی با سه تکرار طراحی و اجرا شد. نتایج نشان داد که تیمارهای آزمایش با کاهش جمعیت باکتری انتهای شاخه گلها، باعث حفظ شادابی و افزایش عمر گلجایی گلها تا سه روز (در تیمار اسانس ppm50 مرزه بختیاری) نسبت به نمونههای شاهد شدند. همچنین با افزایش غلظت اسانس و عصاره هر دو گیاه دارویی، قدرت ضدباکتریایی و عمر گلجایی افزایش یافت. نتایج پس از هشت روز نگهداری نشان داد که بیشترین عمرگلجایی (11 روز)، جذب نسبی محلول گلجایی (8.95 میلیلیتر بر گرم وزن تر در روز) و کمترین جمعیت باکتریایی انتهای شاخه مربوط به تیمار اسانس ppm50 مرزه بختیاری بود. اگرچه از نظر وزن تر نسبی، مواد جامد محلول، محتوایآنتوسیانین، نشت یونی، محتوای مالون دیآلدئید، فعالیت آنزیمهای کاتالاز و فنیلآلانین آمونیالیاز با تیمار ppm50 اسانس آویشن زوفایی فاقد اختلاف معنیدار بود. بنابراین استفاده از غلظت ppm50 اسانس مرزه بختیاری و در مرتبه بعدی غلظت ppm50 آویشن زوفایی بهعنوان برنامهای کاربردی در فرایند نگهداری و بازاررسانی گلهای بریدنی ژربرا توصیه میگردد.

بررسی میزان اثربخشی غلظتهای مختلف اسانس دو گونه مرزه (Satureja khuzistanica Jamzad و S. spicigera (C. koch) Boiss.) در کنترل کنه واروا کندوی عسل
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.635.96.4.1576.1604 کنه واروا از مهمترین آفات کلنیهای زنبور عسل در سرتاسر جهان میباشد، با تغذیه از همولنف لارو زنبور عسل و انتقال برخی ویروسها، خسارت اقتصادی زیادی به صنعت زنبورداری جهان وارد میکند. استفاده از آفتکشهای شیمیایی علیه این کنه باعث ایجاد جمعیتهای مقاوم آفت شده و محصولات کندو را آلوده مینماید. میزان سمّیت تدخینی غلظتهای مختلف اسانس دو گونه مرزه (Satureja khuzistanica Jamzad و S. spicigera (C. koch) Boiss.) بر مرگومیر کنه واروا و زنبور عسل پس از جمعآوری نمونه از زنبورستان در شرایط آزمایشگاهی بررسی شد. کنههای ماده بالغ در دمای 32 درجه سلسیوس، رطوبت نسبی 50% و تاریکی به مدت 5 تا 10 ساعت با غلظتهای صفر، 50، 75، 100، 150 و ppm200 اسانس در چهار تکرار تیمار و برای غلظتهای 20 تا ppm1000 محاسبه شدند. براساس نتایج این آزمایش، مقادیر LC50 تعیین شده برای اسانس مرزه خوزستانی، در بازه زمانی 5 و 10 ساعت پس از شروع اسانسدهی، برای کنه واروا بهترتیب 8/149 و ppm6/78 و برای زنبور عسل کارگر 750.5 و ppm435.8 بود و غلظتهای محاسبه شده برای اسانس مرزه اسپیسیژرا در همان بازه زمانی بر روی کنه واروا، بهترتیب 255.7 و ppm153.6 و برای زنبور عسل کارگر 737.6 و ppm573 تعیین شد. نتایج نشان داد اسانس مرزه خوزستانی و مرزه اسپیسیژرا هر دو با داشتن حداقل تلفات روی زنبور عسل، از قابلیت بالایی برای کنترل کنه واروا برخوردار بودند و مرزه خوزستانی مزیت بیشتری داشت. تجزیه کیفی نشان داد که میزان کارواکرول در اسانس مرزه خوزستانی 93% و میزان کارواکرول و تیمول موجود در اسانس مرزه اسپیسیژرا بهترتیب 43 و 29% بود.

استفاده از روش سطح پاسخ بهمنظور بهینهسازی استخراج عصاره بخش هوایی کرفس (Apium graveolens L.) و ترکیبهای فنولیک و فعالیت آنتیاکسیدانی آن به کمک امواج فراصوت
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.644.96.4.1575.1578 در این تحقیق، اثر زمان (10، 15، 20، 25 و 30 دقیقه) و دمای امواج فراصوت (35، 45، 55، 65 و 75 درجه سانتیگراد)، نسبت اتانول به آب (20، 35، 50، 65 و 80 درصد) و نسبت ماده خشک به حلال (5:1، 10:1، 15:1، 20:1 و 25:1 میلیگرم بر میلیلیتر) بر ویژگیهای فنول و فلاونوئید کل و ظرفیت آنتیاکسیدانی عصاره هیدروالکلی حاصل از گیاه کرفس (Apium graveolens L.) بررسی شد. استخراج عصاره از بخش هوایی کرفس، با روش عصارهگیری هیدروالکلی به کمک امواج فراصوت با بهرهگیری از روش سطح پاسخ انجام گردید و صفات فنول کل، فلاونوئید کل و ظرفیت آنتیاکسیدانی به دو روش DPPH و FRAP اندازهگیری و ارزیابی گردید. براساس نتایج، اعمال زمان 10 دقیقه امواج فراصوت، 55 درجه سانتیگراد دمای امواج فراصوت، نسبت اتانول به آب 80% و نسبت ماده خشک به حلال 5:1 بهترین تیمار بود. در شرایط بهینه، مقدار فنول و فلاونوئید کل بهترتیب 471.12 میلیگرم گالیک اسید در یک گرم ماده خشک و 187.70 میلیگرم کوئرستین در یک گرم ماده خشک و میزان فعالیت آنتیاکسیدانی عصاره به روش DPPH و FRAP بهترتیب 68.48% و 4210.4 میکرومولار Fe+2 در 50 میکرولیتر عصاره بدست آمد. در کل، R2-Adjusted R2 و P-value بالا در برازش مدلها، بیانگر ایدهآل بودن مدلهای سطح پاسخ (RSM) ترسیم شده است. بهعبارت دیگر، بالا بودن این پارامترها نشانگر وجود همبستگی بالای بین مقادیر مشاهده شده و مقادیر پیشبینی شده میباشد.

ارزیابی ویژگیهای رشد و ترکیبهای اسانس در دو گونه دارویی پونهسای برافراشته (Nepeta assurgens Hausskn. & Bornm.) و پونهسای تاجکرکی (N. eremokosmos Rech.)
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.665.96.4.1575.41 دو گونه پونهسای برافراشته (Nepeta assurgens Hausskn. & Bornm.) و پونهسای تاجکرکی (N. eremokosmos Rech.) از گونههای دارویی انحصاری ایران میباشند. این مطالعه در راستای بررسی صفات رویشی، عملکرد اندام هوایی، میزان اسانس و ترکیبهای اسانس و ارزیابی روابط بین آنها در دو گونه مذکور در شرایط مزرعهای انجام شد. آزمایش در قالب طرح بلوکهای کامل تصادفی با سه تکرار، طی سالهای 1396 و 1397 اجرا گردید. نتایج نشان داد که وزن تر و خشک اندام هوایی در گونه برافراشته (بهترتیب 11353 و 2351 کیلوگرم در هکتار) بهطور قابل ملاحظهای بیشتر از گونه تاجکرکی (بهترتیب 2638 و 527 کیلوگرم در هکتار) بود. متوسط درصد اسانس در گونه برافراشته و تاجکرکی بهترتیب 1.60% و 0.64% و عملکرد اسانس بهترتیب 20.16 و 2.21 کیلوگرم در هکتار بود. آنالیز ترکیبهای اسانس بیانگر آن بود که مجموع ایزومرهای نپتالاکتون در گونه برافراشته بهطور متوسط 78.6% و در گونه تاجکرکی 62.8% بود. آنالیز همبستگی بین صفات نشان داد که وزن خشک برگ، وزن تر و خشک کل و قطر بزرگ بوته بیشترین همبستگی را با عملکرد اسانس داشتند. آنالیز رگرسیون گام به گام بیانگر آن بود که تغییرات عملکرد اسانس عمدتاً توسط وزن خشک برگ، درصد اسانس، وزن تر کل، تعداد ساقه و میزان تاج پوشش توجیه میشد. بهطور کلی با توجه به نتایج بدستآمده برتری گونه پونهسای برافراشته نسبت به گونه تاجکرکی قابل ملاحظه بود و میتوان آن را بهعنوان یک گونه ارزشمند برای تولید با مقاصد دارویی توصیه نمود.

ارزیابی تنوع ژنتیکی و گروهبندی جمعیتهای شیرینبیان (Glycyrrhiza glabra L.) با استفاده از تجزیه تشخیص کانونیکی

شیرینبیان (Glycyrrhiza glabra L.) یکی از قدیمیترین و مهمترین گیاهان دارویی تیره Fabaceae است که از 4000 سال پیش برای درمان بسیاری از بیماریها استفاده میشده است. این مطالعه برای ارزیابی و تشخیص منابع تنوع ژنتیکی و فنوتیپی در 22 جمعیت مختلف شیرینبیان در مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه زنجان از سال 1395 تا 1397 اجرا شد. صفات مورفولوژیکی و عملکردی شامل ارتفاع بوته، عرض بوته، طول برگ، عرض برگ، تعداد برگچه، طول برگچه، عرض برگچه، تعداد شاخه جانبی، قطر ساقه اصلی، وزن تر اندام هوایی، وزن تر ریشه، وزن خشک اندام هوایی و وزن خشک ریشه، نسبت ریشه به اندام هوایی، عملکرد اندام هوایی در مترمربع و عملکرد ریشه در مترمربع اندازهگیری شد. در این مطالعه، دو متغیر کانونیکی معنیدار بودند و متغیر کانونیکی اول شامل صفات ارتفاع بوته، عرض بوته، قطر ساقه اصلی، طول برگ، تعداد برگچه و متغیر کانونیکی دوم شامل صفات وزن تر اندام هوایی، وزن تر ریشه، وزن خشک اندام هوایی، وزن خشک ریشه، عملکرد ریشه و عملکرد اندام هوایی بود که بیشترین نقش را در تفکیک جمعیتها داشت. متغیرهای کانونیکی برای گروهبندی جمعیتها به چهار گروه اصلی تقسیم و مورد استفاده قرار گرفتند. نتایج تجزیه تابع تشخیص کانونیکی تفکیک جمعیتها را به گروههای مشابه توسط تجزیه خوشهای تأیید نمود. روش تجزیه تشخیص کانونیکی در ارزیابی تنوع ژنتیکی و شناسایی صفات شاخص، قابلیت خوبی نشان داد.

شناسایی ژنهای مسیر بیوسنتزی اسکلت ترپنوئید در میوه گیاه دارویی هندوانه ابوجهل (Citrullus colocynthis (L.) Schrad.) با استفاده از تکنیک توالییابی RNA
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.691.96.4.1588.65 توالییابی RNA در حال حاضر یک انتخاب مؤثر و پرسرعت برای مطالعه ترانسکریپتوم گونههای گیاهی غیر مدل است که برای شناسایی شبکههای ژنی و الگوهای بیان ژنهای تولیدکننده متابولیتهای ثانویه در اندامهای مختلف گیاهان استفاده میشود. اصلیترین ترکیبها در بافتهای میوه و برگ گیاه دارویی هندوانه ابوجهل (Citrullus colocynthis (L.) Schrad.) را ترپنوئیدها، فلاونوئیدها و آلکالوئیدها تشکیل میدهند. در این مطالعه، ترانسکریپتوم میوه گیاه هندوانه ابوجهل با استفاده از تکنیک توالییابی RNA، بنسازه Iluumina HiSeq2500 اجرا گردید. بعد از کنترل کیفیت با استفاده از نرمافزارهای FastQC و Trimmomatic تعداد 21، 952 و 885 خوانش دارای کیفیت بالا تولید شد و با استفاده از برنامه Evidential-gene بهصورت نوپدید یکپارچهسازی شد که منتهی به تولید 55 و 311 تکژن دارای N50 برابر 927 جفت باز گردید. توالی تکژنهای یکپارچه شده در پایگاه KAAS بارگذاری شد. در مجموع 13، 657 تکژن تفسیر شد که این تعداد در 134 مسیر زیستی قرار گرفتند. مسیر "اسکلت ترپنوئید" با تعداد 93 تکژن از پرتعدادترین مسیرهای شناسایی شده از میان 1، 552 تکژن مسیر بیوسنتز متابولیت ثانویه بود. با بررسی تکژنهای مرتبط با مسیر بیوسنتزی اسکلت ترپنوئید، 29 شناسه ژنی (K) شناسایی شد که تمام ژنهای دو مسیر اصلی بیوسنتزی اسکلت ترپنوئیدی: مسیر سیتوزولی موالونیک اسید (MVA) و پلاستیدی متیل-ارتریتول-فسفات (MEP) از ابتدای مسیر تا تولید ایزوپنیل دیفسفات (IPP) را شامل میشود. شناسایی ژنهای مسیر بیوسنتزی اسکلت ترپنوئید امکان تحقق توسعه تجاری محصول گیاه دارویی که پایهای برای پژوهشهای آینده مربوط به شناسایی مسیرهای بیوسنتزی سایر متابولیتهای اختصاصی، مهندسی متابولیت، اصلاح مولکولی و بهنژادی گیاهان دارویی است را فراهم میکند.

بررسی اثر چهار تنظیمکننده رشد بر عملکرد، رنگدانههای فتوسنتزی و صفات کیفی زعفران (Crocus sativus L.) در شرایط گلخانهای
شناسه دیجیتال (DOR):98.1000/1735-0905.1398.35.704.96.4.1576.1610 بهمنظور بررسی اثر تنظیمکنندههای رشد بر عملکرد و صفات زراعی زعفران (Crocus sativus L.)، آزمایشی در قالب بلوکهای کامل تصادفی در گلخانه تحقیقاتی دانشکده کشاورزی دانشگاه زنجان اجرا شد. تیمارهای آزمایش شامل پرایم بنهها با جیبرلیک اسید (250، 500 و 750 میکرومولار)، سالیسیلیک اسید (700، 1400 و 2100 میکرومولار)، پاکلوبوترازول (50، 100 و 150 میکرومولار) و کلرمکوات کلراید (500، 1000 و 1500 میکرومولار)، هیدروپرایم (آبمقطر) و یک تیمار بدون استفاده از تنظیمکنندههای رشد (شاهد) بودند. در بین تیمارهای بکار برده شده تأثیر جیبرلیک اسید مشهودتر از سایر تنظیمکنندههای رشد بود، بهطوری که بیشترین میزان عملکرد خشک گل و کلاله در تیمار با جیبرلیک اسید و بهویژه در تیمار 500 میکرومولار جیبرلیک اسید مشاهده شد. کمترین عملکرد گل و کلاله مربوط به تیمار بنهها با دو تنظیمکننده کلرومکوات کلراید و پاکلوبوترازول بودند. از سوی دیگر، کاربرد کلرمکوات کلراید و پاکلوبوترازول موجب گردید تا میزان رنگیزههای کلروفیل در مقایسه با دیگر تیمارها افزایش پیدا کند. در مقابل، استفاده از جیبرلیک اسید و سالیسیلیک اسید موجب کاهش کلروفیل شد. تیمار هیدروپرایم اختلاف معنیداری با شاهد نشان نداد. تیمار بنهها با سالیسیلیک اسید موجب افزایش سافرانال (عطر) و کروسین (رنگ) کلالههای تولید گردید. بیشترین اثر روی پیکروکروسین (طعم) در تیمار با کلرمکوات کلراید حاصل شد.
